چکیده

In 1994, in a prospective control trial in cyclosporinetreated, kidney transplant patients, we observed that treatment of a non-specific allograft injury (postischemic reperfusion injury) leads to a significant reduction in the incidence of both specific alloimmune-mediated allograft rejection and chronic allograft failure. From these convincing clinical data, we concluded in terms of an ‘argumentum e contrario’: it is the tissue injury that induces immunity. As from where we stand today in innate immunity research, these early clinical observations can be regarded as the discovery of the existence of a human innate immune system activated by tissue injury and preceding adaptive immunity. Copyright © 2013 S. Karger AG, Basel Current notions in immunology hold that not only pathogen-induced injury but any tissue injury activates the innate immune system leading to infectious/sterile tissue inflammation and preceding adaptive immunity [1–3] . With respect to these modern notions on a fundamental role of the innate immune system in inflammation and adaptive immunity, a peer-reviewed article that dealt with this issue already at an early stage published in Transplantation in 1994 appears to be of interest [4] . In this article, we described the existence of a defense system in humans (without calling it ‘innate immunity’) before first studies on the discovery of the innate immune receptor Toll were published [5, 6] . Interestingly enough, our article appeared just a few months before Matzinger [7] published her famous danger hypothesis which was remarkably in line with our clinical observations. These clinical observations derived from a prospective, randomized double-blind placebo controlled clinical trial that showed a beneficial effect of human recombinant superoxide-dismutase (SOD) on acute and chronic rejection events in kidney-transplanted patients [4] . P-184: The Role of Cell Surface Toll Like Receptors in Endometriosis

Background: Toll like receptors (TLRs) are a major family of innate immune systems which recognize specific pathogen associated molecular patterns (PAMPS)in bacterial, fungi, virus and parasites. Human TLRs comprise a large family of 10 different type proteins that are expressed on various immune cells. Among these receptors, TLR1, 2, 4, 5, 6 and 10 were expressed on the cell surface. TLR2 form...

P-82: The Role of Toll-Like Receptor 2 in Ectopic Pregnancy

Background: The innate immune system is our first line of defense against invading pathogens. Toll–like receptors (TLRs) have been identified as a key role in mediating the function of innate immune system which bridges the gap between innate and adaptive immunity. For example, TLR2 is able to act as a homodimer to recognize lipoteichoic acids of Gram-positive bacteria but as a heterodimer with...

P-17: Expression of Cell Surface Toll-Like Receptors in the Human Male Reproductive Tract

Background: Male infertility refers to the inability of a male to achieve a pregnancy in a fertile female. The root of many causes of infertility is miscommunication between immune and reproductive system. Male reproductive system is very sensitive and vulnerable, infections can hinder maturation and movement of spermatozoa lead to impaired fertility. All species need an immediate reply to the m...

The Efficacy of Toll-Like Receptors in Awakening Dendritic Cell/Natural Killer Cell System for Eradication of Tumors

Natural killer (NK) cells are effector cells of the innate immune system that exert direct cytotoxic functions. Ubiquitously-expressed toll-like receptors (TLRs) have been recognized as one of the major components promoting dendritic cell (DC) maturation, which may induce polarized immune responses beneficial to cancer immunotherapy. TLR-activated NK cells and DCs are prerequisite for robust ac...

P-64: Innate Immune System Importance in Ectopic Pregnancy with Emphasis on TLR4 Gene

Background: In an ectopic pregnancy (EP) the egg does not reach to the uterus, but instead implants somewhere outside it, usually in fallopian tube. In most cases the exact cause of an ectopic pregnancy is unknown but it is often a result of some sort of damage to the fallopian tube. The tube may have become blocked or narrowed by previous surgery or infection. An active immune system need to r...
